Defeating a tough boss/raid for the first time in a game, or finally getting your program to work?

Definitely programming.
Even one tiny tiny tiny tiny mistake will ruin EVERYTHING and you will kill at least half of your lifespan figuring out what went wrong.

Definitely defeating a raid.

Even one tiny tiny tiny tiny mistake will ruin EVERYTHING and you will have to start all over. If this continuously happens, you'll be there for half your lifespan figuring out what went wrong.

Probably most of the voters have never even tried or succeeded on making a program.
Most people here play mmo's though

I'm not really a coder but I do a little from time to time. Having an error you can't fix is infuriating but once you fix it... oh boy

Took me and friends nearly 3 hours to complete Gnomeregan (WoW dungeon) when we were little kids who really sucked at the game, felt so good beating it.

Now we can beat it in roughly 10-15 minutes.(our record is 8- something).

To be fair low level content is so much easier nowadays, especially with heirlooms.

There's nothing quite like getting your HTML and CSS code to finally work properly after numerous upon numerous re-edits and head scratches and confusion, sure beats killing some imaginary dragon with random nerds you never met.
